abstract

objective: embryonic stem cells (escs) are pluripotent cells capable of extensive proliferation while maintaining their potential to differentiate into any cell type. the therapeutic potential of these cells are promising, but in many cases limited by our inability to promote their differentiation. in this study we examined the effect of inducer factors in direct differentiation of mouse escs into a neural fate in suspension culture systems. materials and methods: mouse escs (royan b1) were cultivated in suspension to form embryoid bodies (ebs) within 2 days (2d). they were induced using astrocyte-conditioned medium (acm), retinoic acid (ra, 1μm) and basic fibroblast growth factor (bfgf, 20 ng/ml) for 4 days (2+4d) and were cultured on poly-l-lysine coated dishes up to 5 days (2+4+5d) to differentiate. the expressions of neural specific genes were analyzed by immunocytochemistry and reverse transcription-polymerase chain reaction (rt-pcr). ِِdifferences of means for percentage significance of differentiated ebs were tested by the mann-whitney test and for length and thickness of neural processes by t-test. results: retinoic acid increased the percentage of ebs outgrowth with neural morphology and bfgf had synergistic effect (at least p<0.05), while acm did not influence neural differentiation at all. immunofluorescence analysis revealed the expression of β-tubulin iii in neural cells and that the differentiated neural cells in ra+bfgf group were longer and thicker processes (at least p<0.05). the cells expressed nestin, pax6, nf-m, islet-1, lim1, and hb9. conclusion: the results showed that acm had no effect on neural differentiation from escs, however, ra was in favor of this and that bfgf increased neural differentiation synergistically by mechanisms that remain to be defined.

The Effect of Astrocyte-Conditioned Medium (ACM) and Retinoic Acid on Neural Differentiation of Mouse Embryonic Stem Cells

Purpose: The aim of this research was to study the properties of factors secreted from astrocyte cells in suspension medium in direct differentiation of mouse embryonic stem cells into neural cells. Materials and Methods: Royan B1 mouse embryonic stem (ES) cells were used in this experiment. For differentiation of Es cells into the neural cells, the astrocyte-condition medium (ACM) of mouse fe...

The effect of Fibroblast Growth Factor-2(FGF-2) and retinoic acid on differentiation of mouse embryonic stem cells into neural cells

Introduction: Embryonic Stem (ES) cells as pluripotent cells derived from the inner cell mass of blastula can differentiate to neural cells in vitro and this property is valuable in studies of neurogenesis and in the generation of donor cells for transplantation. In this regard, the propose of this research, was the study of the role of two important factors in the development of neural syst...

the effect of basic fibroblast growth factor on differentiation of mouse embryonic stem cells into cardiomyocytes

introduction: embryonic stem (es) cells are pluripotent cells that derived from inner cell mass (icm) of blastocyst stage and are able to differentiate in vitro into different cells. purpose: this study was initiated to evaluate the effect of basic fibroblast growth factor (bfgf) on differentiation of es cells into cardiomyocytes among with their response to cardiac drugs. methods: the mouse em...

Combined Effect of Retinoic Acid and Basic Fibroblast Growth Factor on Maturation of Mouse Oocyte and Subsequent Fertilization and Development

Objective Many autocrine and paracrine elements that are produced within follicular niche have been the focus of much in vitro maturation research. The present study was carried out to compare retinoic acid and basic fibroblast growth factor (bFGF) efficacy on IVM of mouse oocytes, and their further dual consumption to reach an optimal protocol. MaterialsAndMethods GV oocytes obtained from two-...

Analysis of Promyelocytic Leukemia in Human Embryonic Carcinoma Stem Cells During Retinoic Acid-Induced Neural Differentiation

Background: Promyelocytic leukemia protein (PML) is a tumor suppressor protein that is involved in myeloid cell differentiation in response to retinoic acid (RA). In addition, RA acts as a natural morphogen in neural development. Objectives: This study aimed to examine PML gene expression in different stages of in vitro neural differentiation of NT2 cells, and to investigate the possible role o...

Evaluation of Chronotropic Properties of Mouse Embryonic Stem Cells-Derived Cardiomyocytes After Fibroblast Growth Factor Treatment

Purpose: We investigated the effect of (bFGF) (basic-Fibroblast Growth Factor) on the differentiation of divided cardiomyocytes from mouse embryonic stem cells (ES) and their pharmacological properties. Materials and Methods: The mouse embryonic stem cells (Royan B1) were cultured as 800 cells per 20µl of a hanging drop. After two days, ES cells in each drop aggregated to form embryoid bodies ...
